グリア細胞株由来神経栄養因子(グリアさいぼうかぶゆらいしんけいえいよういんし、英: Glial cell line-derived neurotrophic factor、略称: GDNF)は、ヒトではGDNF遺伝子にコードされるタンパク質である[5]。GDNFは、多くのタイプの神経細胞の生存を促進する低分子量タンパク質である[6]。GDNFはGFRα(英語版)ファミリーの受容体、特にGFRα1(英語版)を介してシグナルを伝達する。
機能
GDNF遺伝子は高度に保存された神経栄養因子をコードしている。組換え型GDNFは培養中のドーパミン作動性神経細胞の生存と分化を促進することが示されており、軸索切断(英語版)による運動神経のアポトーシスを防ぐことができる。GDNF遺伝子にコードされたタンパク質は、ホモ二量体として存在する成熟した分泌型へとプロセシングされる。成熟タンパク質はRETがん原遺伝子の遺伝子産物のリガンドである。GDNFをコードする転写産物に加えて、astrocyte-derived trophic factorと呼ばれる異なるタンパク質をコードする2種類の代替的転写産物も記載されている。この遺伝子の変異はヒルシュスプルング病と関係している可能性がある[6]。
GDNFの最も大きな特徴は、ドーパミン作動性神経[7]と運動神経[8]の生存をサポートする能力である。これらの神経集団はパーキンソン病や筋萎縮性側索硬化症(ALS)の疾患過程で死に至る。また、GDNFは腎臓の発生と精子形成を制御しており、アルコール消費を強力に迅速に抑える効果がある[9]。
GDNFは毛隆起(バルジ)の毛包幹細胞を標的とし、毛包の形成と皮膚の創傷治癒を促進する[10]。
GDNFファミリーリガンド
GDNFは1991年に発見され[11]、GDNFファミリーリガンド(GFL)で最初に発見されたメンバーである。
相互作用
GDNFは、GFRα1(英語版)、GFRα2(英語版)と相互作用することが示されている[12][13]。
治療薬としての可能性
GDNFはパーキンソン病の治療薬としての研究が行われているが、初期の研究では有意な効果は示されていない[11][14][15]。
ビタミンDはGDNFの発現を強力に誘導する[16]。
2012年、ブリストル大学はパーキンソン病患者に対する5年間の臨床試験を開始した。41人の参加者は、薬剤が損傷細胞へ直接到達することができるよう、頭蓋内へのポートの外科的な導入が行われた[17]。GDNFとプラセボの定期的注入による二重盲検試験の結果、GDNF投与群とプラセボ投与群の間でパーキンソン病の症状に関しては有意な差がみられなかったが、損傷した脳細胞に対する効果は確認された[18]。この臨床試験はParkinson's UKの資金提供、The Cure Parkinson's Trust(CPT)の支援の下行われ、CPTの設立者Tom Isaacsも参加者の1人となった[19]。
